ULTRA-RARE, X-LINKED, GLOBAL COMMUNITY
WHEN A DIAGNOSIS HAS NO ROADMAP,
WE BUILD ONE TOGETHER.
UBE2A Deficiency Syndrome affects fewer than 100 known individuals worldwide. Most families receive a diagnosis with almost no guidance, no local specialists, and no clear path forward. The UBE2A Foundation exists to change that — through research, advocacy, and a global community that no family should have to find on their own.
RESEARCHPartnering with scientists, geneticists, and institutions worldwide to advance understanding of UBE2A and accelerate the path toward better care and future therapies.
|
ADVOCACYElevating the UBE2A community voice in healthcare, research funding, and policy spaces — so that ultra-rare does not mean overlooked.
|
SUPPORTConnecting families worldwide with information, resources, and each other — because community is not a bonus. For rare disease families, it is a form of medicine.

UNDERSTANDING THE CONDITION
WHAT IS UBE2A DEFICIENCY SYNDROME?
UBE2A Deficiency Syndrome is an ultra-rare genetic neurodevelopmental disorder caused by a pathogenic variant or deletion in the UBE2A gene, located on the X chromosome at position Xq24. This gene plays a critical role in the body's protein quality-control system — tagging and clearing damaged proteins from cells. When it doesn't work correctly, the consequences reach deep into brain development, neurological function, and daily life.
This is not a condition most doctors have seen before. Most families spend years without a diagnosis — and receive very little information when they finally get one. That gap between what medicine knows and what families are living is exactly what the UBE2A Foundation is working to close.
Commonly reported features include:

Commonly reported features include:
- Absent or severely limited speech — present in nearly all known cases
- Intellectual disability — ranging from moderate to profound
- Seizures and epilepsy — documented in over 50% of published cases
- Hypotonia — low muscle tone affecting development and movementUBE2A: Clinical Profile
- Sensory and behavioral differences — rooted in neurology, not behavior
- Distinctive physical features — facial, skin, and in some cases cardiac
WHY IT PRIMARILY AFFECTS BOYS
ONE X CHROMOSOME. NO BACKUP COPY.
Males are born with one X and one Y chromosome. If that single X carries a UBE2A variant, there is no second copy to compensate. The gene doesn't work — and the full syndrome develops.
Females have two X chromosomes. If one carries a UBE2A variant, the body's X-inactivation system typically silences that copy and uses the healthy one instead. This is why most carrier mothers have no symptoms at all.
This is not just "males are more affected." Virtually every person diagnosed with UBE2A Deficiency Syndrome worldwide is male.
Carrier mothers should be offered genetic counseling. Sisters, aunts, and maternal cousins may also carry the variant and benefit from knowing.

WHAT WE DO
OUR WORK IN THE WORLD.
FAMILY RESEARCH
We are currently conducting a two-year family-reported study in collaboration with the University of Minnesota and the Mayo Clinic, funded by a seed grant from the Masonic Institute for the Developing Brain. Every family who participates directly advances what science knows about UBE2A.
|
GLOBAL COMMUNITY
Our private global support group connects families from across the world — along with researchers who are actively studying UBE2A. When you join, you are not just finding support. You are joining the largest coordinated UBE2A knowledge-sharing effort in existence.
|
RESEARCH
.We bring together leading UBE2A researchers and families to align scientific priorities with lived experience. These roundtables ensure that the questions researchers are asking are the questions families actually need answered.
Upcoming dates are announced within our private community. |
FOR NEWLY DIAGNOSED FAMILIES
JUST RECEIVED A DIAGNOSIS?
START HERE.
Many families receive a UBE2A diagnosis and are told almost nothing — because most clinicians have never seen it before. You are not alone, and you do not have to figure this out from scratch.
